I assume by 'Rothenburg' you mean Rothenburg Ob der Tauber ? I'm also going to assume that you intend to travel by car, though making the journey by train is perfectly feasible. The obvious place to stop on this fairly short journey (less than 3 hours) is Nuremberg, where you should ideally allow at least one full day to explore the fascinating historical centre. Amberg is definitely worth a short detour (or an overnight stop if you wish). Exploring the Gothic Baroque Benedictine monastery complex at Kladruby may also appeal. You could alter (and slightly lengthen) your route to visit Wurzburg en route to Nuremberg. Wurzburg has a lovely historical centre, an impressive Residenz and a couple of excellent and historical weinstuebe: the Juliusspital and the Burgerspital.
