What kinds of things do you like to do?
I don't know if it is "best", but when I was doing it, I started on Route 17 at South Mills (North Carolina) and looked at the Dismal Swamp Canal, continued down to Elizabeth Cityand then skipped over to Knotts Island (North Carolina) and took the ferry over to Currituck.  Then I went over to the Outer Banks  A week in the Outer BanksCorolla with the lighthouse, DuckRoanoke IslandBodie Island Lighthouse took the ferry to , Hatteras and another ferry to Ocracoke and then another ferry to Beaufort (North Carolina)  Take a side trip to New Bern Continuing down the coast to Wilmington  (and maybe a side trip to Bald Head Island.  Then go to Myrtle Beach and Brookgreen Gardens and spend a couple of days in Charleston
You could also start in Cherokee (North Carolina) at the Great Smoky Mountain National Park and take the Blue Ridge Parkway Visitor Center up  to Grandfather Mountain and on up to the Virginia boarder.
