While this drive will technically only take 3+ hours, I would allow at least four hours because of traffic, construction and tolls.  If you go right up I-95 Maryland House  (tolls) to the New Jersey Turnpike (tolls) and through the Holland Tunnel Toll Plaza (tolls) that is the shortest way to New York City. And also the way with the most traffic and congestion and stress.   I hate driving that stretch, but it is the shortest and quickest.

This route bypasses Philadelphia completely.  There are stops on the interstates that you can get gas or eat if you want to do that, but the trip can be done non-stop as long as your bladder holds out. :)

Like others say, totally depends on traffic. But Baltimore to outskirts of New York City is about 3.5/4 hours on an easy drive. Once you get actually into the city....a mile could take 30 minutes.

You can use this page on Trippy to plan a road trip from Baltimore to New York City. On the top right of that page, without any stops, it's about 191 miles with a driving time of 3 hours, 4 minutes.

Traffic and tolls could make a big difference and make the trip longer, but it's definitely doable without stopping. You'd probably want to go around Philadelphia, but if you have time you could also stop in Philly and find somewhere to eat if you're taking a break.
