If time is not an issue, you could go from Louisiana to Natchez, MS and take the Natchez Trace Pkwy through Tupelo all the way to Nashville (or pick it up in Jackson (Mississippi) and end up in Nashville). Natchez Trace Parkway is a scenic drive and features lots of historic sites along the way. In Tupelo you can visit the birthplace of Elvis Presley, tour several museums and Civil War sites, and see bison at the Tupelo Buffalo Park & Zoo. Nashville of course is the center of country music and is famous for the Grand Ole Opry, but also has great food and plenty of other attractions.
From Nashville, take I-40 east through KnoxvillePigeon Forge Gatlinburg and Cherokee (North Carolina) (this route will take you through Great Smoky Mountain National Park. You can go through Maggie Valley to Asheville, but I recommend a detour through the mountain towns of Dillsboro and Sylva. In Asheville you can tour the Biltmore Estate and enjoy more great food. Take Hwy 74 to Chimney Rock State Park and Lake Lure before heading on to Charlotte.
Then on your way back you can take a more southerly route through Atlanta and Birmingham.
