I want to drive on Route 66 as much as possible

How do I plan that trip

Indianapolis (Indiana) Las Vegas (Nevada)

Using Google Maps and the lyrics to "(Get Your Kicks On) Route 66," just enter the towns' names, starting in Chicago, ending in Barstow, CA (skip Winona, AZ), as destinations. (There's a limit to the number of stops you can put into one trip. For the Californa leg, put in Barstow, San Bernadino and Los Angeles.) Don't forget to turn on the "avoid highways" option.
As the Wikipedia article will tell you, most of U.S. 66 has been replaced by limited-access highways (1-40).
