I agree with RosalieAnn, Google Maps is your best friend when it comes to determining a route like this.
But as Michael said - how much time do you have? If you want the fastest route, take the Florida Turnpike as RosalieAnn described.
If you have some time, and are interested in seeing the Florida Atlantic Coast, then follow Florida A1A north from the airport through Boca Raton to Jupiter and then in Fort Pierce (Florida) you will want to get back onto the Florida Turnpike to complete your trip to Kissimmee.
The "scenic Atlantic Coast" route is going to take some time, as it will easily turn your trip into at least a full day's drive. But you are going to pass by some interesting areas, some interesting seafood restaurants, and you will see a lot more of Florida than you would on the Turnpike.

Why do you not use Google Maps for this. When I put in those places, this is what I got (the route has tolls) Get the route from Google maps and pick the one route you want (all of them have tolls) and then print it out.
