Assuming that stl means St. Louis and that charleston means Charleston and not the Charleston which is the capitol of West Virginia, it does seem like Nashville is about half way, but I totally agree with avoiding Nashville because the traffic there is terrible. The one place that I would want to stop in Nashville would be The Hermitage-Home of President Andrew Jackson Asheville is much closer to Charleston.  If I were doing it, I would go through  Louisville and Lexington  Lexington is not quite halfway, but it is a nice little city.  I might also go therough Gatlinburg and then go through the Great Smoky Mountains to Cherokee (North Carolina)

Avoid Nashville. The traffic is terrible. Knoxville is a great place to stay. It is going to be a night visit. Stay at a Hampton or La Quinta. Go to Biltmore in the day time. You will see signs for any hotel you have a rewards membership for. Holiday

The closest major city to the midway point would be Knoxville, TN, but it's not exactly the most interesting place on the planet.  I'd recommend either Nashville or Asheville.  Nashville is a major metropolitan area and Asheville is a small to medium sized city, but both have great food, culture and night life and are great places to spend a few days.
