Hi there! We'll be first time visitors to the U.S. and part of our itinerary will be 4 nights in Cape Cod, mid October driving from Vermont. What are people's opinions for the best town to stay in, perhaps most charming quintessential Cape Cod town with good access to other towns for road trips during the days? Any recommendations on accommodation too? (Research has steered me to Famlouth or Chatham but would love to hear people's ideas!)
I personally would choose the Outer Cape and stay in Chatham (Massachusetts), Wellfleet or Provincetown.
Chatham - Chatham is adorable, very preppy and has a really cute town - you could stay at Chatham Bars Inn
Wellfleet - Personally my favorite town is Wellfleet - The ocean beaches are absolutely gorgeous and the town is cute and a little more low key then Chatham. It is also in the middle of Chatham and Provincetown so you could take day trips to different town on the outer cape.
Provincetown - Really busy in the summer but slows down and is peaceful in the off season. The Red Inn Restaurant is adorable and the food is amazing - For another yummy restaurant I would try Ross' Grill (The view from Ross's Grill is incredible)

Chatham (Massachusetts) would definitely be my first pick and I would try to stay at a B & B, rather than a hotel. Try the Captain's House Inn. It's very New England-y and a great place to stay in the fall.
