If by 'best' you mean fastest/most direct, check the route detailed below. It's around 1100+km (700+ miles), is pretty straightforward with service stations/rest stops, has tolls but avoids driving near or through Paris. Absolute minimum driving time with no stops, traffic congestion, roadworks or navigation issues is 10+ hours.
Driving for 10+ hours is simply not safe so you should plan to break your journey with an overnight stop or, at the very least, a stop of several hours so you can get some proper mental rest (preferably sleep). The geographical (not necessarily time) mid-point of the journey is in the Dijon area.
1. From Calais (France) simply take the A26 southwards, passing through or near NordausquesThérouanneBethuneArrasHargicourtSaint-Quentin and Laon.
2. Follow the A26 around the north-western edge of Reims then continue onto the A4 southwards.
3. Stay on the A4 until just past Les Grandes-Loges then go right to pick up the A26 south again.
4. Stay on the A26 south, passing through or near Mailly-le-Camp and Charmont-sous-Barbuise. South of Daudes go left onto the A5.
5. Follow the A5 east then south, passing through or near BeureyVille-sous-la-Ferté and Marac. Shortly after Marac the A5 joins the A31...make sure you continue south on the A31
6. Follow the A31 south, passing through or near Courcelles-en-MontagneSelongeyChevigny-Saint-Sauveur and Nuits-Saint-Georges
7. The A31 meets the A6 to the north-east of Beaune. Make sure you follow the A6 southwards.
8. Follow the A6 south, passing through or near Chalon-sur-SaoneMacon (Bourgogne) and Villefranche-sur-Saone until you come to the north-east of Lyon where you should turn right onto the A7 south.
9.
