Hi Heather,
Here are my suggestions for a road trip from Boston to Louisville:
1. Drive to Brandywine Creek Campground I assume you will need an RV campground to stay for a night or two. You may want to check out their website. Currently, camping is not allowed. It's about a 5.5 hour drive from Boston, depending on traffic.
2. Visit Valley Forge National Historical Park Longwood Gardens and Brandywine Battlefield Historic Site - all within 20-25 miles from Brandywine Creek Campground. Pennsylvania Dutch and Amish country are about 15 minutes away. 
3. Next stop is Morgantown , WV, about a 4.5 hour drive. There are a couple of RV campgrounds. I found two - Sand Spring Campground and Morgantown Campground.
4. Things to do: visit Laurel Caverns (about 35 minutes from the Sand Springs Camping Area) and go whitewater rafting or rock climbing with Cheat River Outfitters.
5. Drive to Louisville, about a 6.5-hour drive from Morganstown.

It's going to be hot. Go to caves on the way. Mammoth Cave would be great.
