If you had to choose between Mount Tamburine, Hunter Valley or the wine regions near Melborune, which do you think is best. This is based on food, wine and the whole ambiance of the place.

You're going to start an argument with this one, but it probably depends on what wines are your favourite. Australia is vast with different climates in different spots so certain grapes or wine styles are better suited to different regions.
Pinot - cool climate to Tasmania or Yarra Valley in Victoria (Australia) are good. Shiraz - Adelaide wine regions (there are heap there but best known are Barossa/Mclaren Vale. I have been to all three places you mentioned above. If you were choosing a destination for good food/culture, I suggest the Yarra Valley (one hour from Melbourne). The scenery is beautiful and you won't be disappointed by the food.
If you judge a wine region by the average retail price per bottle then the clear winner is the island state of Tasmania. The cool climate wines of Tasmania tend to retail for more than double the Australian average. There are over 200 wineries in Tasmania and 90 cellar door outlets. Tasmania also produces the best whisky in Australia and its compact size makes it a great touring destination. Combine wine with the freshest seafood in Australia. Melbourne wine regions are a must! I would personally visit both the Yarra Valley and the Mornington Peninsula. Both regions are not only known for their great wine but also great local fresh produce. I live on the Mornington Peninsula so I am a big advocate when it comes to our wine. There is also a large variety of sightseeing and activities available on the Mornington Peninsula. A few places worth visiting is the beautiful Fort Nepean, the Cape Schanck Lighthouse Reserve or you may even consider taking a swim with the dolphins in Port Phillip The options are endless.

Definitely hunter valley. But another great wine area is Margaret river in Western Australia!
