Depends what you mean by best. Here is the easy way (less walking, fastest):

Take the train ("Peru Rail") to Aguas Calientes from one the following stops: Urubamba, Ollantaytambo and I believe Cuzco (takes a few hours). Buy tickets online at http://www.perurail.com/.

Once you arrive in Aguas Calientes, take the bus to Machu Picchu. It takes about 30 minutes. The bus ticket counter is a 5 minute walk from the train stop (follow the crowd).

The hard way is to take Inca trail. You have several options and it varies from 2-7 days.

The best way is to arrive by way of the Inca Trail ! It's a challenging...but extremely rewarding...4 day hike that will have you hiking over some seriously beautiful mountain passes in the steps of the ancient Incas. Many tour companies offer trek packages that include everything - food, accommodations, transfers and porters and guides on the trail. I recently did a "G Adventures" tour that included Cuzco, the Sacred Valley and an all-inclusive trek on the trail. Everyone in my group - a mix of all ages from many countries - made the trek safely and successfully and were rewarded with the most amazing view of Macchu Picchu at sunrise.
